My basic Pathogenic route is:

  1. Choose a Pathogen whose movement I can control.
  2. Build one dependable forward attack.
  3. Spend DNA on an Evolution that creates useful space.
  4. Use Cores to fix movement, recovery, Range, or a missing connection.
  5. Clear short valuable branches before each boss.
  6. Stop exploring when another unknown room is more dangerous than useful.
  7. Preserve Health, Stamina, and movement for the Stomach and Heart.

The normal organ route is:

Skin → Intestines → Stomach → Liver → Lungs → Heart

What Pathogenic Actually Tests

Pathogenic is not only a damage-stacking roguelite.

Every run asks me to solve four connected problems:

  • how the body moves;
  • where each weapon points;
  • which Organelles are connected;
  • whether the Build can survive long enough to apply its damage.

My weaker runs usually had one impressive number and one obvious structural problem.

Examples include:

  • strong frontal damage with no rear protection;
  • several active abilities with no reliable charge trigger;
  • a wider Evolution without better movement;
  • a melee Build that could not approach ranged enemies;
  • a projectile Build that failed as soon as an enemy reached the body.

The order that works better for me is:

movement → firing lane → close-range protection → synergy → more damage

I do not need every category perfected in the Skin.

I need each one to improve before the later organs begin combining environmental hazards, mixed enemies, and multi-form bosses.

Choose a Pathogen by Movement First

Pathogenic has seven playable Pathogens:

  • Bacterium;
  • Helminth;
  • Fungal Spore;
  • Amoeba;
  • Nanobot;
  • lil collector;
  • Diatom.

I check movement rules, body shape, special slots, starting traits, and loadout progression before choosing a Pathogen.

For a first normal clear, I prefer Bacterium.

Its front is easy to read, and its body works naturally with one forward ranged weapon, side protection, and a later projectile-bounce setup.

The other Pathogens change what I value:

  • Helminth: controlled melee with projectile protection;
  • Fungal Spore: several connected weapons without overfilling the body;
  • Amoeba: slots and body development before specialization;
  • Nanobot: compact modular connections and careful Health management;
  • lil collector: automatic, orbiting, or bouncing attacks that tolerate changing angles;
  • Diatom: separate damage and defensive roles between two players.

I do not choose only by the starting weapon.

A strong opening attack is less useful when I cannot keep it pointed toward the escape route.

The First Rooms: Build a Foundation

The Skin does not need to produce the final Build.

My opening target is:

  1. one attack that works without a long charge;
  2. enough DNA for a useful Evolution;
  3. Cores for a meaningful shop choice;
  4. enough Health to survive an unfamiliar enemy;
  5. one movement or control improvement before the body becomes large.

Clear the Threat Before Collecting Resources

DNA and Cores can fall while enemies are still active.

I use this order:

  1. Remove the nearest ranged threat.
  2. Create a safe forward lane.
  3. Kill enemies beside or behind the body.
  4. Recover enough Stamina to move again.
  5. Collect visible DNA and Cores.
  6. Check the map.
  7. Choose the next room.

I do not cross an enemy firing lane merely because a resource is glowing.

I also do not leave loose DNA indefinitely after the room becomes safe, because dropped resources can disappear.

DNA, Cores, and Plasmid Do Different Jobs

DNA Changes the Current Body

DNA advances the current run’s Evolution.

An Evolution may provide:

  • a new body shape;
  • more external slots;
  • more internal space;
  • different firing directions;
  • another Stamina container;
  • additional damage or movement options.

I choose the Evolution that fixes the current Build. More total slots are not automatically better when the new body is harder to turn.

I take a larger form when:

  • the new slots point into useful lanes;
  • I already have Organelles ready to fill them;
  • Movement Speed or Stamina is acceptable;
  • the next route has enough space for the body.

I take Stamina or preserve the smaller body when:

  • the current weapon stops firing too soon;
  • the larger form would remain mostly empty;
  • the body already turns slowly;
  • a boss is close;
  • movement is currently keeping the run alive.

Cores Buy Immediate Run Value

Cores are used for shops, rerolls, rewards, and other run decisions.

I spend them when the purchase fixes something specific:

  • Range;
  • Movement Speed;
  • Stamina;
  • Health;
  • side or rear protection;
  • a missing connection;
  • a compatible Overcharge trigger.

I save them when:

  • the item cannot fit;
  • the trigger cannot activate;
  • the Build already clears rooms safely;
  • the purchase only duplicates an existing attack;
  • another accessible shop has better known value.

Unused Cores are not wasted when the alternative is a bad purchase.

Plasmid Improves Later Attempts

Plasmid progression happens between runs.

Each Pathogen has its own longer-term progression and loadout objectives.

I prioritize the problem that repeatedly ends that Pathogen’s runs:

  • movement when the body cannot reposition;
  • starting capacity when the opening layout is too restricted;
  • Health or Armor when one mistake ends the run;
  • damage when ordinary enemies survive long enough to surround me;
  • rerolls or shop advantages when the Build repeatedly lacks compatible pieces.

Plasmid progression does not replace learning the movement system, but it gives that movement more room to work.

Build Organelles as a Network

The editor contains several connected roles.

Weapons Create the Attack

Weapons include:

  • projectiles;
  • lasers;
  • melee strikes;
  • orbiting attacks;
  • acid;
  • lightning;
  • continuous beams.

Direction matters as much as damage.

A powerful weapon in a side-facing slot may spend most of the room attacking empty space.

Infusers Improve Connected Weapons

An Infuser does not automatically affect the whole body.

I follow the visible connection lines and check which weapon actually receives the effect.

One well-connected attack is usually better than several unsupported weapons.

Mitochondria and Triggers Power Active Effects

Active Organelles may charge through:

  • successful attacks;
  • kills;
  • entering combat;
  • finishing a room;
  • finishing without losing Health;
  • sustained Overcharge.

The trigger must match the next encounter.

A kill trigger may work well while exploring but begin empty against a boss.

A successful-attack trigger is usually more dependable against one large target.

The Three Build Directions I Use Most

The full combinations and Tier List are in the Best Builds Guide.

For general planning, I use three recognizable structures.

Ricochet Laser Core

I use:

  • one laser or rapid projectile weapon;
  • Elastosome or another bounce effect;
  • Range;
  • one movement improvement;
  • one close-range answer.

This is my safest general ranged direction, especially on Bacterium and developed Fungal Spore runs.

Frozen Blade Shell

I use:

  • Freeze;
  • Kinetosome or another orbiting defense;
  • a stable ranged weapon;
  • projectile control;
  • enough Stamina to reposition.

This works well when mixed enemy groups or narrow Stomach rooms keep closing the firing lane.

Armor Reactor

I use:

  • a functioning Armor source;
  • a repeatable Overcharge engine;
  • dependable forward damage;
  • defensive control.

The important Hub-level rule is that an unfinished Armor engine should not replace Health, movement, or a weapon that already works.

When I Buy Health Instead of Damage

The simplest shop mistake is buying another attack when the run is already one hit from ending.

I buy Health when:

  • the boss is close;
  • one normal hit could end the attempt;
  • I am still learning the next pattern;
  • the body is difficult to move;
  • the existing damage already clears ordinary enemies.

I buy damage when:

  • Health and Armor are stable;
  • enemies survive long enough to surround me;
  • the new item strengthens the existing firing lane;
  • the item has a compatible slot and trigger;
  • movement and control are already covered.

The correct purchase addresses the reason the run is taking damage.

It is not automatically the safest-looking or highest-rarity reward.

When I Reroll, Recycle, or Leave

I Reroll When the Offers Cannot Solve the Current Problem

Useful reasons include:

  • every offer is offensive while movement is failing;
  • the main weapon needs Range;
  • Health is critical;
  • the Build needs a compatible Infuser or trigger;
  • an unlock objective specifically requires a reroll.

I do not reroll repeatedly in search of a theoretical perfect item.

I Recycle When an Organelle Has No Job

I recycle it when:

  • no valid slot exists;
  • it cannot connect;
  • the direction is unusable;
  • the effect duplicates something already covered;
  • it blocks a more important piece.

I keep it when the next confirmed Evolution can realistically make it useful.

I Leave When the Build Is Already Ready

I stop changing the layout when:

  • the main weapon reaches the target;
  • the body can retreat;
  • a close-range threat has an answer;
  • the boss trigger works;
  • another change would require relearning every direction.

A stable Build is allowed to remain imperfect.

Explore Known Value, Not Map Completion

The rooms are procedural, so I use a decision rule rather than a fixed path.

I explore when:

  • the branch is short;
  • the route is safe;
  • Health is stable;
  • I still need movement, recovery, or a key connection;
  • a known shop is only a few Cores away;
  • the Boss Build is incomplete.

I turn back for known value:

  • Health;
  • Armor;
  • DNA that immediately reaches an Evolution;
  • an opened shop;
  • a visible reward;
  • a fast-travel point;
  • an unlock objective.

I stop exploring when:

  • the remaining branch is long and unknown;
  • Health has become fragile;
  • environmental pressure is closing the route;
  • the Build already passes the boss check;
  • another reward would require rebuilding the layout;
  • the next transition becomes one-way.

The rule I reuse across the game is:

Known value is worth backtracking for; hypothetical value is not.

The Normal Organ Route

Skin

The Skin is where I learn the current body.

I focus on:

  • one forward firing lane;
  • the first useful Evolution;
  • a shop or reward route;
  • basic movement and Dodge timing.

I do not expect the final Build here.

I only need a foundation that reaches the Intestines without losing every defensive resource.

Intestines

The Intestines begin punishing one-direction Builds.

Enemies can reach the side or rear while ranged attacks continue from ahead.

This is where I add:

  • close-range protection;
  • side or rear coverage;
  • projectile control;
  • enough Stamina to keep firing while moving.

A second frontal gun is not always the correct answer.

Stomach

The Stomach is the first major route, environment, and boss check.

It introduces:

  • corrosive acid hazards;
  • narrow routes;
  • mixed enemy pressure;
  • a closing death zone;
  • the two-form Ossified Macrophage boss.

Difficulty Customization includes an option that disables the closing Stomach death zone while leaving the enemies, acid, and boss intact.

I use that setting when the timer prevents me from learning the organ itself.

Use the Stomach Guide for acid timing, procedural routing, shop-return decisions, Pathogen advice, and both boss forms.

Liver

The Liver tests whether the Build can keep scaling without becoming unreadable.

By this point, I stop adding unrelated weapons.

I improve:

  • the main damage chain;
  • movement;
  • survivability;
  • Range;
  • reliable triggers.

The Build should become clearer as the run continues, not more complicated.

Lungs

The Lungs make movement impossible to ignore.

Airflow can push the body, change the firing lane, and trap a wide form.

I watch the flow before spending all my Stamina fighting against it.

Before leaving, I collect known rewards that still matter.

The Heart circulation route becomes one-way.

Heart

The Heart ends with the Atrioventricular Node.

The fight combines:

  • limbs controlling parts of the arena;
  • summoned enemies;
  • laser and projectile patterns;
  • recovery pickups;
  • a transformation into a second form.

I enter with:

  • one sustained ranged attack;
  • an answer for enemies reaching the body;
  • movement beyond the starting value;
  • enough Stamina for the transformation;
  • a trigger that works against one boss.

Use the Heart Boss Guide for named pattern descriptions, limb priority, phase-two movement, Pathogen advice, and Antibiotics adjustments.

Nanobot Is a Separate Progression Objective

Nanobot requires:

At least 8 Armor held at the same time

Armor already lost to damage no longer counts toward the current total.

The unlock does not require defeating the Heart.

Use the Nanobot Unlock Guide for:

  • the recommended starting Pathogen;
  • verified Armor sources;
  • the seven-Armor danger point;
  • procedural bad-luck recovery;
  • the final one-way route;
  • Nanobot’s later Build identity.

What Changes After the First Heart Clear

The first normal Heart victory is not the end of account progression.

It can open or advance:

  • Endless Mode;
  • Antibiotics difficulty;
  • Pathogen-specific Heart achievements;
  • Plasmid progression;
  • loadout objectives;
  • the longer route toward the Brain.

I do not begin Antibiotics merely because one Heart clear succeeded.

I first want a repeatable normal clear where:

  • movement is already upgraded;
  • one limb can be removed without becoming trapped;
  • adds do not shut down the main firing lane;
  • enough recovery remains for phase two;
  • the Build still works while active abilities recharge.

I then combine later Antibiotics clears with unfinished Pathogen-specific Heart achievements wherever possible.

Experiments Change the Rules of the Run

Experiments are not simply stronger versions of a normal route.

They may change:

  • map length;
  • item-room count;
  • visibility;
  • weapon restrictions;
  • starting layout;
  • charging rules;
  • mirrored slots;
  • time pressure.

I read the full Experiment rule before choosing a Pathogen. A restricted weapon pool or shorter timer changes what is valuable from the first room.

One visible Experiment setup added:

  • 15 more rooms;
  • two additional item rooms;
  • a three-minute closure timer.

Other rules included Lights Out, Weapons Only, random starting Builds, and modified active-Organelle charging.

The Achievements Guide organizes all 15 Experiment achievements alongside organ bosses, loadouts, Antibiotics, and the Brain.

The Boss Check I Use Before Committing

Before entering any major boss, I ask:

  1. Can the main attack reach the target?
  2. Can I stop an enemy that reaches my side?
  3. Can the body retreat without rotating through danger?
  4. Can the active effects charge during this fight?
  5. Can I survive more than one mistake?

When one answer is no, I look for the nearest short route that can fix it.

When every answer is yes, I stop shopping and enter.

Common Mistakes That Ended My Runs

Filling Every Slot

More Organelles made the layout harder to understand without fixing its weak direction.

Taking a Larger Body Before Movement

The new slots were useful, but the wider form could not reverse quickly enough.

Buying Damage at Critical Health

The Build already cleared rooms. It needed enough Health to survive another mistake.

Ignoring Trigger Conditions

Powerful active effects remained empty because the Build could not perform the required action.

Rebuilding Immediately Before a Boss

A higher damage number did not compensate for unfamiliar controls and firing directions.

Entering the Boss as Soon as I Found It

A short nearby branch often contained the Health, movement, or connection the Build still needed.

Exploring After the Build Was Ready

The final unknown room sometimes consumed more Health than its reward could replace.

For the first clear:

  1. Start with this Hub.
  2. Open the Best Builds Guide when the first useful Organelles appear.
  3. Use the Stomach Guide before the third major wall.
  4. Read the Heart Boss Guide before entering the one-way Heart route.

After the first clear:

  1. Complete the Nanobot Unlock Guide when the eight-Armor objective becomes realistic.
  2. Use the Achievements Guide to combine Pathogen clears, Antibiotics, Experiments, loadouts, and Brain progression.

The House Always Wins FAQ

Does disabling the Stomach death zone also remove acid hazards? +

No. The setting removes the closing-zone pressure, but corrosive acid, ordinary enemies, room hazards, and the Ossified Macrophage still remain.

Do I need to defeat the Heart to unlock Nanobot? +

No. Nanobot unlocks as soon as the current run holds at least eight Armor. Once the notification appears, the unlock remains saved even if the later Heart attempt fails.

Does the first Heart clear immediately unlock the Brain? +

No. A normal Heart clear can open Endless Mode and Antibiotics progression, but the route beyond the Heart remains closed until additional account progression is completed.
